Research Interests
His laboratory focuses on the pathophysiology, cell biology and genetics of pancreatic disorders. Landmark contributions include:
- identification of the pancreatic acinar cell as the primary injury site in pancreatitis,
- discovery of transient pancreatic duct obstruction and missorted endocytosis as initiating events in gallstone pancreatitis,
- elucidation of calcium-mediated protease activation and lysosomal enzyme involvement,
- demonstration of magnesium’s ability to block trypsin activation (now in clinical trials),
- characterisation of immune-cell (macrophage/NLRP3 inflammasome) roles in onset and severity of pancreatitis,
- genetic risk stratification (ABO-B, FUT2) and metabolomic biomarker signatures distinguishing pancreatitis from cancer,
- establishment of the exocrine pancreas as a key regulator of intestinal microbiota.
Scientific Recognition
Professor Lerch’s work has been honoured with numerous international awards, including:
- American Pancreatic Association Lifetime Achievement Award (2021)
- George-Palade-Prize in Cell Biology (2018)
- Lifetime Achievement Award of the European Pancreatic Club (2016)
- Elected Member of Academia Europaea (2016)
- Honorary Fellow of the Royal College of Physicians and Surgeons of Glasgow (2001)
