Roger A. Greenberg is the J. Samuel Staub, M.D. Professor, Director of Basic Science at the Basser Research Center for BRCA, Program Leader in the Breast Cancer Program at the Abramson Cancer Center, and Director of the Penn Center for Genome Integrity. Affiliated with the Perelman School of Medicine at the University of Pennsylvania, he leads a lab focused on DNA repair, genome integrity, and their implications in cancer biology and immune signaling.
Research Interests: Dr. Greenberg’s work centers on the molecular mechanisms of DNA repair, particularly BRCA1 and BRCA2-dependent homologous recombination in breast and ovarian cancers. His team investigates telomere maintenance via alternative lengthening of telomeres (ALT), DNA damage-induced immune activation, and chromatin dynamics in DNA repair. Key areas include ubiquitin signaling, PARylation, and chromatin remodeling as they relate to PARP inhibitor sensitivity and cancer therapy.
Publication Trends: His recent articles highlight advancements in ubiquitin-dependent DNA damage recognition, ALT telomere repair, and DNA damage-immune crosstalk. The work spans Molecular Biology, Genetics, Oncology, and Immunology, with a focus on chromatin modifications, enzyme complexes like ALC1 and BRISC, and clinical translation of DNA repair inhibitors.
Grants & Collaborations: Dr. Greenberg's lab collaborates with institutions like the University of Leeds and has developed first-in-class BRISC deubiquitinating enzyme inhibitors. His work is supported by clinical trials (e.g., NCT06525298) and fellowships awarded to lab members.
Labs & Teams: The Greenberg Lab at Penn includes a team of research specialists, graduate students, and postdoctoral researchers, with administrative support from Laura Murillo. The lab is part of the Penn Center for Genome Integrity, advancing translational research in DNA repair deficiencies.
