Karl Schmitz is an Assistant Professor in the Departments of Biological Sciences and Chemistry and Biochemistry at the University of Delaware, College of Arts and Sciences. His research focuses on bacterial proteases, particularly Clp proteases in Mycobacterium tuberculosis, aiming to uncover their role in bacterial survival and to develop novel antibacterial strategies.

Research Interests:
Schmitz's laboratory investigates the structure, function, and regulation of ATP-fueled proteases in bacteria, with a primary focus on Clp proteases. These enzymes are essential for protein quality control and survival under stress conditions. Using biochemical, biophysical, and structural approaches, his team seeks to understand how these proteases recognize and degrade specific substrates, and how their activity can be targeted pharmacologically.
Current projects include:
- Understanding Clp protease assembly and disassembly dynamics
- Identifying physiological substrates and selectivity mechanisms
- Developing high-throughput screening assays for antibacterial compound discovery
Funding and Impact:
In 2024, Schmitz received a five-year, $1.88 million R01 grant from the NIH/NIAID to study the role of phosphoarginine modifications in M. tuberculosis and their interaction with Clp proteases. This work has significant implications for global health, especially in regions where tuberculosis remains a major threat.

Schmitz mentors both graduate and undergraduate students, including doctoral candidates in Biological Sciences and Chemistry. His collaborative environment fosters interdisciplinary training in microbiology, biochemistry, and chemical biology.
