Dirk Remus is an Associate Professor in the Molecular Biology Program at the Sloan Kettering Institute, Memorial Sloan Kettering Cancer Center, and affiliated with the Weill Cornell Graduate School of Medical Sciences. His research focuses on the molecular mechanisms of DNA replication, particularly how replisomes function in the context of chromatin and respond to replication stress.
His research interests include DNA replication, genome stability, chromatin dynamics, replication stress, checkpoint signaling, and cryo-EM structural analysis of replisomes. The Remus Lab studies how multi-subunit replisome complexes navigate obstacles such as RNA polymerases, DNA damage, and non-canonical DNA structures during chromosome replication. Using reconstituted yeast and human systems, the lab investigates the regulation and structure of replisomes to understand replication fork stalling and recovery mechanisms.
The recent publications (2020–2023) highlight a strong focus on R-loop dynamics, replisome-checkpoint interactions, and structural mechanisms of replication proteins. Key themes include transcription-replication conflicts, G-quadruplexes, CMG helicase regulation, and the 9-1-1 DNA damage clamp.

Dr. Remus leads an active research laboratory and mentors trainees in molecular and structural biology. His lab’s work is supported by institutional and possibly external grants, and it contributes to the development of novel cancer therapeutic strategies by targeting DNA replication machinery. He collaborates with structural biologists and cancer researchers across the Tri-Institutional community.
The Remus Lab is actively involved in studying replication stress mechanisms, programmed replication fork barriers, chromatin replication, and DNA damage checkpoint signaling, utilizing biochemical reconstitution and cryo-EM approaches.
