Anne Goldfeld is a Professor in the Department of Pediatrics at Harvard Medical School and leads the Goldfeld Laboratory at Boston Children's Hospital. Her research spans transcriptional regulation, HIV pathogenesis, and tuberculosis immunology with a focus on host-pathogen interactions and innate immunity.
Her laboratory pioneered the paradigm of cell type- and stimulus-specific inducible eukaryotic gene regulation through distinct enhanceosomes and higher-order chromatin structure, particularly studying TNF gene expression in T cells, B cells, macrophages, and fibroblasts. Key discoveries include identification of SNPs marking primate ancestry in the TNF promoter, characterization of distal enhancers and matrix attachment regions in the TNF locus, and elucidation of NFAT protein roles in transcriptional activation. Her HIV research uniquely examines co-infection with Mycobacterium tuberculosis, analyzing viral isolates across global HIV subtypes to identify conserved regulatory elements.
Her groundbreaking clinical work established the CAMELIA trial in Cambodia determining optimal timing of antiretroviral therapy for HIV/TB co-infection, which transformed treatment guidelines worldwide. The laboratory also identified the first gene associated with tuberculosis susceptibility and characterized IL-10-producing T regulatory cells in infectious disease.
Recent publications demonstrate expanding research into SARS-CoV-2 pathogenesis, revealing mechanisms of monocyte infection and inflammasome activation, while continuing HIV/TB research with novel antiviral and antimicrobial strategies including nitazoxanide repurposing. Her work consistently bridges basic molecular mechanisms with clinical applications in resource-limited settings.
