معرفی
Thomas J. Silhavy is the Warner-Lambert Parke-Davis Professor of Molecular Biology at Princeton University, where he has been a founding member of the Department of Molecular Biology since 1984. His research focuses on the molecular mechanisms of outer membrane biogenesis and envelope stress response in Gram-negative bacteria, particularly Escherichia coli.
Education:
- B.S. in Pharmacy, Ferris State College (summa cum laude, 1971)
- M.S. and Ph.D. in Biological Chemistry, Harvard University (1974, 1975)
- Postdoctoral training with Jonathan Beckwith, Harvard Medical School
Research Interests: Silhavy's work centers on understanding how bacterial cells assemble complex structures like the outer membrane, a critical permeability barrier. His lab investigates protein targeting, signal transduction, and the cellular machinery involved in outer membrane protein (OMP) and lipopolysaccharide (LPS) assembly. This research has implications for developing novel antibiotics that target the outer membrane, potentially overcoming current resistance mechanisms. His lab uses a combination of genetics, biochemistry, and bioinformatics to dissect these pathways.
Recent Publications Trend: His recent publications highlight a sustained focus on the β-barrel assembly machine (Bam), envelope stress responses (e.g., σ^E pathway), and the physical and regulatory aspects of outer membrane biogenesis. A notable trend is the exploration of small-molecule inhibitors targeting BamA, offering new therapeutic avenues. His work also increasingly incorporates systems-level analyses, such as protein turnover and phase separation in membranes, reflecting a comprehensive approach to bacterial physiology.
Scientific Awards and Honors:
- Member, National Academy of Sciences (2005)
- Associate Member, EMBO (2008)
- ASM Lifetime Achievement Award (2016)
- NIH MERIT Award (1999)
- Novitski Prize for Creativity (2008)
- President’s Award for Distinguished Teaching, Princeton (1993)
- Editor-in-Chief, Journal of Bacteriology (2012–present)
- Jack Kenney Award for Outstanding Service (multiple years)
Advising and Grants: Silhavy has advised numerous graduate students and postdoctoral fellows, contributing significantly to training the next generation of scientists. His long-standing support from the NIH, including a MERIT award, underscores the impact and continuity of his research program. He has also been deeply involved in academic service, notably as Editor-in-Chief of the Journal of Bacteriology, where he has championed scientific integrity and equity, as seen in his co-authored statements on diversity in microbiology.
Labs and Research Teams: The Silhavy Lab at Princeton, located in the Thomas Laboratory, is a leading research group in bacterial molecular biology. The lab fosters a collaborative environment, combining classical genetic approaches with modern biochemical and systems biology techniques to address fundamental questions in cell envelope biogenesis.
