João Mamede, PhD is an Associate Professor in the Department of Microbial Pathogens and Immunity at Rush Medical College, Rush University. His research focuses on understanding the intricate interactions between viruses and their hosts, with particular emphasis on HIV infection mechanisms in various immune cells.

Dr. Mamede's research program centers on the molecular mechanisms of HIV infection in CD4 T-cells, macrophages, and dendritic cells. His laboratory employs advanced techniques including long live-cell fluorescence imaging, high and super resolution fixed immunofluorescence, and various biochemical and biophysical assays to study viral entry, uncoating, and early replication events. A significant portion of his work investigates how host factors, particularly Interferon Stimulated Genes (ISGs), either promote or restrict HIV infection. His research has important implications for understanding viral pathogenesis and developing novel antiviral strategies.
Analysis of Dr. Mamede's recent publications (2022-2025) reveals a strong focus on HIV-1 infection mechanisms, particularly the role of viral capsid in evading host immune responses. His work increasingly incorporates advanced imaging techniques to track viral particles in real time within host cells. There's also a notable expansion into studying HIV's effects on the nervous system using cerebral organoid models, as well as investigations into the intersection of HIV research with other viral pathogens like SARS-CoV-2.
Dr. Mamede currently leads a research team comprising Stephanie Gambut (Research Lab Tech), Charia McKee (PhD Candidate), James Szczerkowski (Research Scientist), Janet Zayas (PhD, Instructor), and Li Zhang (PhD, Postdoctoral Fellow). His laboratory has secured funding through NIH grant K22 AI140963 for "Direct live-cell visualization and quantification of interactions between infectious HIV particles in primary target cells" and a CHEETAH Center Development Award for studying early steps of the HIV replication cycle.
