John C. Burnett, M.D., is a Professor of Medicine and Physiology at Mayo Clinic in Rochester, Minnesota, where he serves in the Department of Cardiovascular Medicine. He holds the Marriott Family Cardiovascular Research Professorship and is recognized as a Distinguished Investigator at Mayo Clinic. His research focuses on the endocrine function of the heart, particularly the role of natriuretic peptides in cardiorenal homeostasis, with direct applications in heart failure, hypertension, and myocardial infarction.
- Marriott Family Cardiovascular Research Professor, Mayo Clinic (2007–present)
- Distinguished Investigator, Mayo Clinic (2006)
- Member, American Society for Clinical Investigation (1990–present)
- Member, Association of American Physicians (1994–present)
Dr. Burnett earned his M.D. from Loyola University Stritch School of Medicine in 1974, followed by residency and fellowship training at Mayo Clinic, including specialized training in cardiovascular disease and nephrology. He has led a continuously NIH-funded research program that has pioneered the development of designer natriuretic peptides, resulting in three novel therapeutics currently in clinical trials, seven patents, and the founding of three biotechnology companies: Nile Therapeutics, Anexon, and Zumbro Discovery.
His research interests span cardiovascular medicine, natriuretic peptide biology, novel drug development, and biomarkers. His laboratory’s landmark 1986 Science paper demonstrated elevated ANP in heart failure, establishing the foundation for BNP as a diagnostic biomarker. Current work includes dual receptor-activating peptides, small molecule modulators of guanylyl cyclase, and innovative delivery platforms.
The most recent 15 articles (2023–2025) reflect a strong emphasis on novel peptide therapeutics, oxidative stress modulation, genetic underpinnings of cardiomyopathy, and dual-pathway targeting in heart failure. These works highlight a trajectory toward precision medicine, multi-target therapies, and translational innovation in cardiorenal disease.
- National Institutes of Health (NIH) – multiple grants (1985–present)
- National Heart, Lung, and Blood Institute (NHLBI) – active projects on peptide therapeutics and small molecule discovery
- Mayo Clinic – institutional support and training programs
Dr. Burnett has advised and collaborated with numerous researchers, including S.J. Sangaralingham, X. Ma, and P.M. McKie. He has held leadership roles including Director for Research at Mayo Clinic (1999–2004) and continues to chair key research committees. His work bridges basic science, translational development, and clinical application, significantly impacting the understanding and treatment of cardiorenal diseases.
His laboratory has developed innovative platforms for chronic peptide delivery, including subcutaneous, gene-based, and oral systems, and continues to advance natriuretic peptide-based diagnostics and therapeutic strategies for resistant hypertension and heart failure.
