معرفی
David E. Cobrinik serves as Associate Professor of Research in Ophthalmology and Cancer Biology at the University of Southern California, with primary laboratory affiliation at Children's Hospital Los Angeles (CHLA). His research program investigates retinal development mechanisms and their relationship to ocular diseases, with specialized focus on retinoblastoma pathogenesis and novel diagnostic approaches.
Dr. Cobrinik's research centers on the cellular origins of retinoblastoma, where his laboratory established that cone photoreceptor precursors serve as the tumor cell-of-origin. His work demonstrates how cone-specific signaling pathways collaborate with pRB loss to drive tumorigenesis, revealing new therapeutic targets. Current investigations focus on three key areas: (1) developmental signaling pathways that sensitize retinal cells to Rb loss; (2) molecular events converting normal retinal cells to malignant retinoblastomas; and (3) targeting vulnerabilities in pRB-deficient cone precursor circuitry. Additionally, his team pioneers human pluripotent stem cell models to recreate retinal development in vitro, enabling unprecedented study of human retinal biology and disease mechanisms.
Analysis of Dr. Cobrinik's publication record (2020-2024) reveals dominant themes in liquid biopsy development for retinoblastoma, particularly aqueous humor-based detection of tumor DNA and biomarkers. His laboratory has established clinical utility for aqueous humor liquid biopsy in diagnosis, prognosis, and treatment monitoring. Significant work also explores retinoblastoma's cellular origins in cone precursors and molecular mechanisms involving MYCN/MDM2 signaling, alongside stem cell-derived retinal organoid models for studying development and disease.
