Charles K. Barlowe holds the James C. Chilcott 1920 Professorship and serves as Chair of the Department of Biochemistry and Cell Biology at the Geisel School of Medicine, Dartmouth College. He leads an active research laboratory investigating fundamental mechanisms of intracellular protein transport.
His research focuses on molecular mechanisms of ER-Golgi transport, employing yeast and mammalian models with biochemical, genetic, and microscopic techniques. Key areas include compartment identity maintenance, cargo receptor functions in directional protein trafficking, and connections between ER quality control defects and human diseases like cystic fibrosis, Alzheimer's, and cholesterol disorders. The lab actively investigates how luminal environments regulate cargo-receptor binding and how golgin proteins organize Golgi compartments.
Recent publications demonstrate sustained productivity in cargo receptor mechanisms, particularly the Erv41-Erv46 complex in retrograde transport and Erv14's role in COPII vesicle formation. This work reveals how cargo receptors discriminate folded/misfolded proteins and maintain organelle identity amid bidirectional traffic.
