William Klein is a Professor in the Department of Neurobiology at Northwestern University's Weinberg College of Arts & Sciences. His research focuses on the molecular mechanisms underlying Alzheimer's disease, particularly the role of amyloid-β (Aβ) oligomers in neurotoxicity and disease progression. He holds a PhD from UCLA and has been a pioneer in elucidating how Aβ oligomers disrupt synaptic function, leading to cognitive decline.
Education:
- PhD in Neuroscience, University of California, Los Angeles (UCLA)
Research Interests: Dr. Klein's work spans therapeutic development, diagnostics, and understanding the etiology of Alzheimer's. His team investigates:
- Neurotoxic effects of Aβ oligomers on synapses and memory mechanisms
- Development of monoclonal antibodies for clinical trials targeting Aβ oligomers
- Ultrasensitive diagnostic assays and imaging techniques (PET/MRI) for early detection
- Metabolic factors (e.g., diet-induced diabetes) contributing to Aβ oligomer buildup
- Structural biology of Aβ oligomers using cryo-EM and mass spectrometry
Collaborations & Affiliations: Klein collaborates globally, including with institutions like the Federal University of Rio de Janeiro, Osaka University, and the Karolinska Institute. He is affiliated with Northwestern's Cognitive Neurology Alzheimer's Disease Center (CNADC) and the Chemistry of Life Processes Institute (CLPI).
Future Work: Current efforts include refining drug candidates (e.g., sabirnetug) for clinical trials, advancing non-invasive diagnostics, and exploring insulin signaling pathways to counteract Aβ toxicity. His lab also uses novel models like brain organoids and induced pluripotent stem cells (iPSCs) to study disease mechanisms.
