Sara Buhrlage serves as Associate Professor of Biological Chemistry and Molecular Pharmacology at Harvard Medical School, Harvard University, where she leads the Buhrlage Lab for Cancer Chemical Biology.
Her research pioneers pharmacological validation of deubiquitylating enzymes (DUBs) as therapeutic targets through first-in-class inhibitor development. Focusing on 'undruggable' cancer targets involving protein-protein interactions, her work centers on targeted degradation of oncoproteins via the ubiquitin-proteasome system. Key methodologies integrate peptidomimetic approaches, DUB inhibitor libraries, and validation of degradation mechanisms for cancer treatment.
The Buhrlage Lab maintains an integrated DUB-centric platform featuring novel chemotype libraries, biochemical/chemoproteomic assays for compound profiling, and multidisciplinary expertise spanning synthetic chemistry, structural biology, computational chemistry, and functional annotation to accelerate clinical translation of DUB-targeted therapeutics.
