Herbert Kim Lyerly serves as the George Barth Geller Distinguished Professor of Immunology and holds joint appointments as Professor of Surgery and Professor of Pathology at Duke University School of Medicine. He maintains critical affiliations with the Duke Cancer Institute, Duke-Margolis Center for Health Policy (as Core Faculty), Duke Regeneration Center, and Duke Global Health Institute, directing translational initiatives through the Surgical Sciences Center for Applied Therapeutics and Environmental Health Scholars Program.
His educational foundation includes an M.D. from UCLA (1983) and distinguished scholarly recognition as a Life Member of Clare Hall, University of Cambridge (2022-2030), with prior Visiting Fellowships at Cambridge in 2019 and 2022. This international engagement complements his deep institutional roots at Duke.
Dr. Lyerly's research program centers on overcoming immunotherapy resistance in breast and prostate cancers through innovative vaccine platforms targeting oncogenic drivers like ESR1 mutations and androgen receptor variants. His lab pioneers spatial analysis of tumor heterogeneity using mouse models and develops ultrasound-enhanced delivery systems (e.g., PULSE technology) to improve intratumoral immunotherapy penetration. Concurrently, he investigates environmental health disparities, particularly air pollution's impact on cancer outcomes and multimorbidity.
Analysis of his 15 most recent publications reveals accelerating innovation in three domains: (1) Mechanistic studies of antibody-drug conjugates like trastuzumab deruxtecan, focusing on payload release and immunomodulation; (2) Next-generation vaccine strategies using mRNA-LNP platforms to target treatment-resistant mutations; and (3) Advanced imaging techniques (hyperspectral, spatial) for mapping tumor-immune interactions. These efforts consistently bridge basic discovery and clinical application.
His leadership in sponsored research is evidenced by 10 major concurrent grants totaling over $50 million, including a 10-year NIEHS Environmental Health Program (2019-2029), DoD-funded breast cancer initiatives through 2028, and Susan G. Komen-supported work on obesity-tumor interactions in TNBC. These projects integrate basic science, clinical trials, and population health approaches.
Dr. Lyerly directs the Surgical Sciences Center for Applied Therapeutics, which develops novel drug delivery platforms, and co-leads the Environmental Health Scholars Program training next-generation researchers in pollution-cancer linkages. His work with the Duke-Margolis Center actively shapes health policy frameworks for cancer care delivery and clinical trial diversity.
