Sarah E.F. D'Orazio, PhD is a Professor and Vice Chair for Education in the Department of Microbiology, Immunology, and Molecular Genetics at the University of Kentucky. Her laboratory is located in the Medical Sciences Building (Rooms MS417 for office and MS417 for lab) where she leads research on Listeria monocytogenes pathogenesis and host immune responses.
Dr. D'Orazio earned her Ph.D. from the University of Miami School of Medicine and completed postdoctoral training at Harvard Medical School. Her research focuses on understanding host susceptibility and resistance to Listeria monocytogenes infection, with particular emphasis on foodborne transmission routes. She developed a novel mouse model for oral transmission of Listeria that closely mimics all phases of human disease: (1) ingestion of contaminated food, (2) gastrointestinal infection phase, (3) systemic spread in susceptible vs. resistant strains, and (4) brain dissemination.
Her laboratory currently has two NIAID-funded projects: one investigating intracellular growth niches for Listeria in the ileum and colon, and another studying how neurotropic strains spread directly to the brain via axonal migration without high blood titers. Her work has significant implications for understanding foodborne pathogens, particularly as Listeria contamination in ready-to-eat foods remains a persistent public health concern with a high fatality rate (~30%) among high-risk populations including neonates, pregnant women, the elderly, and immunocompromised individuals.
Analysis of her recent publications (2016-2024) reveals a consistent focus on Listeria monocytogenes pathogenesis, with particular attention to host immune responses, bacterial dissemination mechanisms, and genetic factors influencing susceptibility. Her research spans immunology, microbiology, and infectious disease, with increasing emphasis on neurotropic strains and brain invasion mechanisms in recent years. The work employs sophisticated mouse models, cellular immunology techniques, and molecular microbiology approaches to dissect host-pathogen interactions at multiple tissue sites.
Dr. D'Orazio leads an active research laboratory with current members and alumni, contributing significantly to the understanding of foodborne pathogens and host immunity. Her work bridges basic science with important public health implications, particularly regarding food safety and vulnerable populations at risk for severe listeriosis.
