Kenneth L Rock serves as Chair and Professor in the Department of Pathology at UMass Chan Medical School. He also holds significant positions at the T.H. Chan School of Medicine (Department of Pathology, Division of Basic Pathology) and the Morningside Graduate School of Biomedical Sciences (Immunology and Microbiology Program and MD/PhD Program).
Dr. Rock earned his BA in Biology from Washington University in St Louis and completed his MD at the University of Rochester School of Medicine in 1978.
His research focuses on the fundamental mechanisms of immune surveillance, particularly how MHC class I molecules display foreign antigens to the immune system. His laboratory investigates both cross-presentation (where antigens are acquired from dying cells by professional antigen-presenting cells) and direct presentation pathways. A major contribution of his work has been identifying endogenous adjuvants like uric acid (particularly monosodium urate crystals) that are released during cell death and stimulate immune responses. This research has significant implications for understanding tumor immune evasion and developing novel immunotherapies.
Dr. Rock's recent publications (2017-2025) consistently focus on antigen presentation mechanisms, particularly MHC class I pathways, and their implications for cancer immunology. His work spans from fundamental immunological mechanisms to translational applications, with increasing emphasis on how tumors evade immune detection through alterations in antigen presentation pathways and transcriptional regulation of immune-related genes.
Dr. Rock actively mentors graduate students through the GSBS program, offering rotation projects that explore immune surveillance mechanisms including danger signals, innate immune responses, dendritic cell biology, and antigen presentation pathways. His laboratory provides students with comprehensive training in immunology, experimental design, and technical skills while contributing to advancing scientific knowledge.
His laboratory focuses on the molecular mechanisms of immune surveillance, particularly antigen presentation pathways, regulation of immune responses through endogenous adjuvants, and how these processes can be harnessed for therapeutic applications in cancer and infectious diseases.
