Houfu Guo

Houfu Guo, PhD, is an Assistant Professor in the Department of Molecular and Cellular Biochemistry at the University of Kentucky College of Medicine. His research focuses on collagen-modifying enzymes and post-translational modifications (PTMs) and their roles in cancer, fibrosis, and connective tissue disorders.

Dr. Guo earned his Ph.D. and M.S. from the University of Kentucky and a B.S. from the University of Science & Technology of China. His lab investigates how lysyl hydroxylase 2 (LH2) and other collagen PTMs regulate the biomechanical and biochemical properties of the extracellular matrix, particularly in diseased stroma. The research employs structural biology, biochemical assays, animal models, and small molecule screening to develop therapeutics targeting LH2.

His work spans collagen biology, extracellular matrix signaling, enzyme kinetics, and cancer stroma remodeling. Recent publications highlight discoveries in LH2 inhibition, collagen glycosylation in lung cancer, and stromal heterogeneity. Trends in his research show a strong emphasis on molecular mechanisms of fibrosis and metastasis, with increasing translational focus on drug development.
