Claudio Punzo, PhD is a Professor in the Department of Ophthalmology at the University of Massachusetts Medical School School of Medicine. He serves as Principal Investigator of the Punzo Lab, which focuses on developing treatments for photoreceptor degenerative diseases. His research is primarily funded by the NIH and patient foundations, with a strong emphasis on translational approaches to preserve vision in retinal degenerative conditions.
Dr. Punzo's research interests center on understanding the mechanisms of photoreceptor loss in genetic inherited diseases like Retinitis Pigmentosa (RP) and age-related diseases such as age-related macular degeneration (AMD). His lab has pioneered work showing that metabolic dysregulation and nutrient starvation contribute significantly to cone death in RP. A key finding from his research demonstrates that activation of the mTORC1 pathway can significantly prolong cone survival in RP models, providing a promising therapeutic target. His work also extends to AMD, where his lab has developed a novel mouse model that develops characteristic features of geographic atrophy.
Analysis of Dr. Punzo's publication record reveals a strong focus on retinal metabolism, gene therapy vectors, and therapeutic targets for photoreceptor preservation. His recent work (2023-2025) has increasingly focused on novel AAV capsids for improved retinal delivery, oligonucleotide therapeutics, and molecular mechanisms underlying AMD pathogenesis. The research trajectory shows a progression from fundamental mechanisms of photoreceptor death toward developing specific therapeutic interventions.
Dr. Punzo actively mentors students and postdoctoral researchers in his lab, with documented graduations such as Aditya Venkatesh in 2016. The Punzo Lab maintains partnerships with both academic and industry collaborators to advance therapeutic development. The lab utilizes a range of techniques including mouse modeling, viral vector delivery, metabolic analysis, and molecular biology approaches to address fundamental questions in retinal degeneration.
The Punzo Lab operates within the supportive ecosystem of the Horae Gene Therapy Center at UMass, which provides specialized resources for gene therapy development. The lab actively recruits postdoctoral associates, research associates, graduate students from the Morningside Graduate School of Biomedical Sciences, and welcomes part-time students, summer students, and volunteers interested in vision research.
